If the Tangle Bloom isn't dead after that, just hit it with everything you've got, and it will fall quickly. Open by summoning if you have any Djinn built up, then cast any fire-elemental spell at your disposal. Now seems like the perfect time to do so. However, what they were doing was building up your standby Djinn count, allowing you to summon a crazy-powerful beast. Also, during your romp through the dungeon, Isaac and Garet were casting their Djinn spells seemingly at random. Any fire elemental spell, such as Fireball, will trash him pretty quickly.
